I am using single tape for Enterprise vault backup. If tape get full and it will ask second tape to continue the same backup job. can we configure E-mail notification tape alert to user

Logon to OpsCenter / Manage / Alert Policies / Add / Device, Mount Request.
If you want the alert to be emailed, then in OpsCenter, also configure an email recipient, and configure SMTP target details.
Netbackup will never ask for a tape for backup jobs.
If you do not have free tapes available at the same pool or at the scratch pool, the backup will fail (error 96).
backup exec has this functionality. (to ask for a tape for backup)

But we are not using tape library, I have single tape drive LTO 4 and my EV data size in 2. 5 TB
FYI - Backups to standalone drives in NetBackup will wait for a tape.

Suppose my one tape is full and it will ask to second tape for the same backup job. How to know backup job asking to second tape, if is there any alert notification send to user so user will understand backup job asking second tape.

Sorry for that. I have to work with standalone tapes for more than 10 years.
I do not think that Opscenter have such email alert. But I may be wrong again.
You can write a script that checks for pending requests using the "vmoprcmd -d pr" command and if there is a pending request to send email to the operators.

Even there are no logs getting generate in windows event viewer.
