One of the machine picked by the SEPM 12.X as infected one ,I checked the status of that machine locally(SEP Client) which is green saying "your computer is protected ,no problem detected", why it is conflicting

What does it show in the Risk log? Run a full scan to see if it is clean.

Nothing in the Risk Logs regarding the instances picked by the SEPM today although they are in the still infected and newly infected status,
the scan has come out with 0 detections

get the risk log from SEPM for the client and check for the drive /device(USB) where infection was found.

Sep version 12.1
and the infection came out of a file inside the local machine not from any external drive

"your computer is protected ,no problem detected" this means SEP is running fine. It does not tell about the risk.
no risk found for client from SEPM risk logs?

the version used is old one, can you try upgrading SEPM to the latest version .
