Have up installed all the latest updates for Backup Exec? What OS you are trying to backup?
Enable SGmon log before running the job and check for errors in the log.
To enable SGmon log refer to the below link
http://www.symantec.com/docs/TECH4986

Can you confirm the version of Backup Exec you are currently using?
If it is BE 2012 or earlier, you may want to refer to the following tech doc: http://www.symantec.com/docs/TECH136185

I am using Symantec Backup Exec 2010 R3.
I have read this doc : http://www.symantec.com/docs/TECH136185 but I don't find the path to exclude.
In the log, I don't have error about a file which cause a problem.
I think this message is important. Have you got an idea :

Enable "Shadow Copies" of all the volumes and set it to no limit.
Retry the backup. If it still fails, are you able to run a successful backup using the native Windows backup utility ?
