What version of BE and Domino are you using?  Also, are you using the Domino Agent?

The Version of BE is 2010 R2 Sp1 and we are running Domino 8.5.2. Yes, the Domino Agent is installed. The Domino Agent backsup only the notes folder. The remaining folders of the server are backed up by the backup job.

Please check if the right database is being backed up that is if the lotus domin database is being backed check in the selection or the backup job. it might be possible that another database (with .ntf extension )is being considered as the lotus notes database so check the name. Mayuresh

Dear Mayuresh Thule,

the Lotus Agent is configured to backup a Template file (*.ntf). There is only one Template File in our Notes folder. Isn't it possible to backup a *.ntf File with the Domino Agent ?

The Lotus Domino agent keys off of the database file extensions when locating them for backup.  When creating a backup job, if you select non-database files that have .NSF or .NTF extensions under the Lotus Domino agent, Domino server returns an error when Backup Exec requests to open them during backup.  We've commonly seen this with some postscript(?) files on the system drive of Windows servers cause this error.  As long as you limit the backup selections beneath the Lotus Domino icon to locations containing only Domino databases normally this error does not occur.
