Hi,
We are facing this problem with Backup Exec 2010 R3 and SBS2008 Exchange server.
Exchange server is not logging any errors but Backup Exec Server is:
Backup Exec Alert: Job Failed
(Server: "F-STX200") (Job: "Backup 00015") Backup 00015 -- The job failed with the following error: Cannot extract mailbox messages from the Exchange backup. Review the job log for more information.
And Backup Exec gives this error:
Error: e0000395 - Cannot extract mailbox messages from the Exchange backup. Review the job log for more information.

This seems to be exactly same problem as you have documented in support pages: http://www.symantec.com/docs/TECH68132
I've already tried removing agent and license and after rebooting i added exchange license and installed remote agent again, but this didn't work.
I also tried to create new backup job, but this didn't work either...
What could suddenly cause this problem when this job has been running nicely for months?

Hi, same Probleme here (BE 2010R2 on SBS 2008).
The Problem started appearing occationally after R3 inplace-Upgrade (from 2010 non-R2).
Error Code is also e0000395 . and detailed Error description is "-1032 The file cannot be accessed because the file is locked or in use. ".
This looks quite similar (except for the different error Code which I never saw before) to an old Case with similar Problems with 2010 and 2010-R2 until a Patch was released: https://www-secure.symantec.com/connect/forums/error-0xe0000393-v-79-57344-915-cannot-extract-mailbox-messages-exchange-backup .
I tried to excluse the Temp Dir as stated in TECH128115 but I doubt this will help.
Switching AOFO off (which shouln't matter for 2008 Installations anyway) let the Job rnu succesfully for one Day but the Error returned one Day after.
If I so a single Backup with just the Exchange Database (complete copy only as in the Jobs), it runs succesfully.
RAM usage is below 50% (18 GB RAM, about 60% free atm) and there is also plenty Disk space available.

I have this same problem in a Hyper-V server. Fully patched and updated Backup Exec 2010 R3 running on Server 2008 parent. SBS 2011 running as a VM with Windows, Exchange and Hyper-V agents. I get this message about once a week and I am wondering if it has anything to to with our Trend Micro Worry Free Business Security Advanced installed on the SBS VM.
I looked at http://www.symantec.com/business/support/index?page=content&id=TECH128115 that indicates,
'This issue is resolved by excluding the Backup Exec temp directory C:\Temp\Backup Exec on the Exchange server from Anti-Virus active scan.'
I do not have a C:\Temp\Backup Exec folder on either the parent server (Hypervisor) or rhe SBS 2011 VM. Any suggestion? There is a configuration setting in Trend Micro Worry Free Business Security Advanced that will aloow me to exclude folders. Exchange is already excluded.
