The error indicates your backup server 'Backup1' cannot connect to the backup exec database which exists on 'DB1'. I would assume from this that when backup exec was setup a SQL instance was created on 'DB1' as opposed to creating a local MSDE instance on 'Backup1' .

Check that the Sql services on 'DB1' are running. Open Enterprise Manager and verify that the BEDB database is running.

When did the error occur- what changed?

The error just occurs every now and then in the event log.
I can read the DB for restores etc, but it comes up every now and then.
DB services are started and the database is running. Maybe nothing to worry about.

It could be also a network issue with packets being lost.
Try fixing the LAN connection by forcing the use of 100 MB and Full Duplex on the NIC and switch configuration.
