Hello!
When I start a Backup of the Informationstore my two active Mailbox DBs are moved to one MBX Server.
My enviroment:
All Server win2k8R2
2x Exch2k10sp1 CAS/HT
2x Exch2k10sp1 MBX with 2 active DBs and 2 inactive DB copys (Activation Preference set)
1x BackupExec 2010R2
svc_backup= Domain Admin = AD user for BackupExec.
The backup job ist a full backup. Without backup the DAG works correct.
The Informationstore backup is successful at all time successful.
Some events that occurs by the backup proccess from the MBX Server:

The solution was: disable Advanced Open File and configure a preferred server list.
However I have always set to backup only the informationstore.

Disable tcpoffloading on the NIC and see if that helps ..

To check whether the backup account has the proper role assignments you can run the following from the 2010 Exchange Powershell.
Get-Mailbox {username} | ft name,roleassignmentpolicy
To change it if it is not set run
Get-Mailbox {username} | Set-Mailbox –RoleAssignmentPolicy “Default Role Assignment Policy”
