We are facing issue with user few users as below.
Connection from DLO clients to DLO Server fails with the error "Failed to load configuration settings" and starting the DLO client prompts the error "Failed to load configuration settings" .

Try to disable the AV to see if it is causing the issue?
If the problem persists, I advise logging a case and have Support check this.

If you could post the DLOClient.log file from C:\users\<username>\appdata\local\VERITAS\DLO\.settings it will have the hint for the exact cause of the issue. When it's saying Unable to Load configuration settings it could be due to various reasons, to list the few.
1. DLO Server is not reachable
2. NUDF is not reachable
3. DLO Database server is not reachable
4. Misconfiguration of Profile
5. AV blocking the communication
Also you can try to run the DLODiagnostics Utility on the Client where you are getting the error and that would pretty much give you the hint to look at the right place.
You can run this DLODiagnostics Utility from Start | All Programs | VERITAS DLO | DLO Diagnostics Utility
OR
Go to C:\Program Files(x86)\VERITAS\VERITAS DLO\DLO\ DLODiagnostics Utility. Run Scan Now and this will try to run some standard checks and will point out the area for the cause of the issue.
