2. if we run the manual backup of any client will it affect the schedule of that client the next time it will run how it would be calculated?

1. Retry option start the failed backup again within the backup window. For example, if your have retry option set to 2, backup will be attempted on its  own for 2 times within the backup window (if backup fails withing backup window) after 30 min interval (default setting, you can change this). This option works only for the scheduled backups and not manual or user started backups.

2. If you run manual backup of any client it will affect the next run if the Schedule is based on the Frequency.
