when i will reboot drive ,is there any issues in running backup ?

To answer your question: 

If you have running backups on the drive, then the backups will fail.
The OS and NBU will lose connectivity to the drive.

Question - why would you want to reboot a drive while a backup is running?
If the backup is really running, then it seems that there is no problem?

Several time TLD has been down.

So,i have to do manually updrive many times.

Thats why i want to reboot.

Chances are that a reboot of the server or the tape drive(s) will not fix your problem. 

You need to find out why drives are being DOWN'ed. 

There are many forum posts and Veritas Technotes with possible reasons, e.g
3 x I/O errors in 12 hours on the same tape drive
Certain hardware TapeAlert errors from the tape drive firmware
Tape drives unable to unload media for various reasons (incorrect mapping, tapes manually loaded in robot slots, faulty tape that is stuck in drive, etc)
etc... etc...

To know the exact reason for drives being DOWN'ed, add VERBOSE entry to vm.conf on all media servers, then restart NBU Device Management service (with no backups running).

NBU will now log additional device-related info to Windows Event Viewer System and Application logs. 
Next time a drive goes DOWN, the exact reason will be logged in Application log. 
Hardware errors will be logged in System log. 

In the meantime, ensure that bptm log folder exists on all media servers (no restart necessary).
Also check Tape Log report for errors. 
You can also check   ...\NetBackup\db\media\errors file to see errors logged against media and/or drives.
