iSCSI can be used as disk storage provided it is fully supported by VMware (as per the documention that I have referred you to in my previous post).
If you want to use iSCSI as dedupe storage, be sure to read up in NetBackup Deduplication Guide about using iSCSI disk.
Are you referring to a client or a media server that is the backup host?
The only requirement for a backup host is that NBU client software must be installed.
It can also be a media server or even the master server that is used for all kinds of backups.
There is no need for a backup host to be dedicated to VMware backups only.

As long as you are aware of the fact that only disk-based storage can be used with VM media server...
