Can LTO3 backup tape from BE12 or 12.5 restore on BE2010 LTO5 ?

Yes should be there...
But if you have truncated catalogs, the directories and files cannot be restored untill the media is recataloged.

Yes, it can be done.
In general LTO 5 tape drives have backward read/write compatibility to LTO-4 media and backward read compatibility to LTO-3 media.
And BE 2010 will be able to read the media which was written by earlier versions of BE. You need to have the catalogs of that media to make the restore process easy. Otherwise, you need to catalog the media. It will take a lot of time.

The BE 2010R2 is inplace upgrade from BE 12 .
So the media catalog should be still inplace on the BE 2010.

I have test the restoration this afternoon and it work fine for the file restore.
As long the tape are expired , i have to rerun the "catalog media".
How long will it take to recatalog a lto3 size of data?

There is no such measures exactly.
But in my experience once it took four and half hours to catalog LTO3 media.
Some times it may take more than that also...
