How can I create a large initial replica for a remote site in an infrastructure with slow WAN connections?

Your best plan would be to backup the data from the remote site and restore them to your local target system.

Would this approach be appropriate in the scenario where a very large amount of data was added to a system that was already being replicated?

this depends on your environment, resources and staff. You are more on the lines of a backup/restore and also a cloning type of scenario. You can also make the replica from the remote site by putting the hardware there and replicating the data so that you dont lose any information over the WAN.
