There are quite a bit of guides there for tuning on Windows. As well as optimum BackupExec settings.

Looking at some performance guides for the DD640 (I can't share them as they are for resellers only)
The DD640 over a single GbE connecction over CIFS, can do 80MB/s reads. Over 10GbE, can do 120MB/s over CIFS. It's much faster if you use other protocols like NFS, BOOST or VTL. VTL being the fastest.
How are you connected? You should be using LACP for the two onboard GbE connections. You should be on the latest DDOS5.1.
I would suggest you also search my.datadomain.com to read up on LMDD and IPerf. Both are tools to test actual througput from Media server to DD box. This is the ONLY way to measure throughput accurately for the DD unit. I'm not sure how you are getting your numbers currently.
