I believe you do need to have the SQL Server agent installed to do a complete backup of the database, though many people recommend getting SQL to do a backup of the database itself, and then backing up the flat file with backup exec.

If you do choose to use Backup Exec to backup the live database you will need to ensure you have patched your Backup Exec installation, I think it's patch 16 you need which enables support for SQL 2005 though I could be wrong on the patch number.

It's Hotfix 6 you need to add SQL 2005 support to 10d. See http://www.backupexecfaq.com/faq/problems/sql-2005-with-be-10.html

There's some (rough) information on backing up SQL (including the method Keith mentioned) at http://www.backupexecfaq.com/faq/concepts/backing-up-microsoft-sql-server.html
