Please explain your setup a bit more.

Are you running Exchange 2010 ?
Is your BE Media server a 64 bit 2008 server ?
Is Exchange running in a DAG configuration ?

You have installed MAPI/CDO on all DAG members ?

You have installed the Exchange 2010 management tools on the BE media server ?

Also check the permissions of the serviceaccount following this article:

Try running the job from active copy ( active node ) of the DAG. Usually it is caused due to replication writer / exchange writer stability issues.

On the Backup job properties,

Go To : Settings --> Microsoft Exchange --> High Availability Server --> Backup from Active Copy only

You may also check the writer on the passive node by running the following command from command prompt :

c:\>vssadmin list writers

Are you able to ping to the DAG name ? and IP address ?

Also check if you have the managment tools installed on the Media server which should be of same or greater version than exchange.

What happens if you directly try to backup the database from server and not from DAG node ?

Check if the databases are visible under DAG node in backup selections. If they are not listed in backup selections under DAG node then follow: http://www.symantec.com/docs/TECH161618
