I have one machine as the server 4 windows 2000 sp4 and the rest are Windows 2008 r2. Windows 2000 is connected to the Tape Backup is a way around this problem, however.

Tape drive has to be connected to the media server(server where backup exec is installed)
If you are installing BE on a VM than make sure OS detects the tape drive.

As prevously advised, you need to attach the tape drive to the media server. There is no way around this.
Also, Symantec does not advised the use of a VM for the media server because there will be problems accessing real devices.

If you are using a Hyper-V based VM then there is not SCSI pass-through - as such BE cannot support a tape drive inside it because the native environment does not support it.
Also to Support Exchange 2010, Backup Exec must be installed on a 64 bit server that can have the Exchange tools installed on it.
As such you need to connect your tape drive to a physical Windows 2008 R2/x64 server and install Backup Exec on that server to backup your environment.

Your first issue is that the media server runs Windows Server 2000, which as per the TN below, is not a supported OS to run Backup Exec 2010 R3. Check the SCL below:

You have 1 of 2 ways to fix this:
1. Upgrade the OS of the media server to ast least Server 2003 R2 (Server 2008 R2 is better if the architecture supports it!)
