Files still grow after hotfix

"Random file on Backup Destination Server grows to 256 GB during a continuous backup job in Continuous Protection Server (CPS) version 12.5 or Continuous Protection Server (CPS) version 2010."

Files randomly grow even after the hotfix (cpspatch.exe) has been installed

I have backup exec 2010 build 13.0.418.8101 running on windows server 2008 standard.

Is this BE 2010 or BE 2010 r2 sp1

As per the link when you update the hotfix the file version should be updated to 13.0.420.3701 & as per your above post the file version in your case is 13.0.418.8101

This is BE 2010 not r2.  The link you sent is referring to r2.  The bellow link says that it will be version 13.0.418.8101

Then please upgrade to BE 2010 r2 & CPS 2010 r2 & install the hotfix given above & see if that helps
