Hello Jose,
This is a very generic error : 0xe0009585 - Unable to open a disk of the virtual machine
the cause to this error could be as follow:
1. Permission on the logon account.
2. Tranport mode used to access the VMDK.
Please confirm the following:
Check if the account used to backup the vmdk had admin privileges in the Vcenter. If not assign admin role to the account.
If the VMDK are SAN mounted, the SAN luns on which the VM resides need to be presented to Media Server.
REF: http://www.symantec.com/docs/TECH63541
If the SAN luns are not presented, you can use the NDB mode to backup the virtual machines.
The tranpsort mode can be set under "virtual machine" configuration in the job.
If the following does not help create a job for the VM, with the following debug enabled in the registry so that we find the exact error:
Debug: http://www.symantec.com/docs/TECH63926 Follow step 1 to 5
Thanks.

Unable to access your datastore through your VM proxy server/off host backup server. Check the transport mode it might be SAN or NBD which is visible to your datastore.

Which transport mode are you using in the VMware settings of the job?
