Windows 2008 R2 Enterprise SP1, Backup Exec 2012 - fully patched, MS Hotfix 979612 and 982383 installed.
Windows firewall an AV software disabled (Server and Client)
Intel Xeon E5520 2,27GHz, 4 Cores
24 GB RAM

If not, client side deduplication will not work.

I have the same problem:
One backup to disk job, but two duplicate to tape jobs at the same "level". One is running every sunday, the other one on every first sunday of every month.

Overwrite protection 7 days is not a good idea as the overwrite protection is "set" to the media after the job finished. So it will be protected for one week + job running time
I never used Tapes with Backup Exec 2012, but if it is like in 2010 then you can do it this way:
