Can i enable a new store partition whilst backups are running on current partitions

What do you mean by 'enable a new store partition' ?

I have just created a new store partition and left it as ready as my backups are running, can i close the current active whilst the backups are running or is it best to let backups finish before i close the current active partition and active the new.
