Which version of NetBackup are you using and which Operating System is the client (2003 /2008 etc)?
There are many bugs in earlier versions that can cause this sort of issue as well as the client itself.
What do the clients that fail do? (file servers, SQL , AD etc.)

NBU Version 7.5.
Windows Server 2008R2
FileServer

Is that 7.5 GA or have you patched?
Is it a standalone server or a cluster node? (http://www.symantec.com/docs/TECH199089)
There is also a MS Hotix that you may need http://www.symantec.com/docs/TECH176793
