As per Compatibility List lee posted, NetBackup 6.5.4 or later support vSphere 4.0. See page 9.
Support for VMware vSphere 4.0
NetBackup 6.5.3 and 6.5.4 support vSphere configurations with VCB 1.5 Update
1 and Converter 4.0.1 as minimum requirements.
To use NetBackup 6.5.3 with vSphere, additional engineering binaries (EEBs) are
required. Please contact NetBackup support and request the EEBs associated
with Etrack 1633379. Installation instructions can be found in the following
Symantec tech note:

But NetBackup 6.5.x support VCB backup only. To configure vStorage API backup, NetBackup 7.x is required.

NetBackup 6.5 supports ESX Server 3.0, 3.0.1, 3.0.2, 3.5, abd 3i. Here is a link to a doc that specifies support for NetBackup in virtual environments, and applies to NetBackup 6.x, http://www.symantec.com/business/support/index?page=content&id=TECH64542.

6.5GA(6.5.0) does not suppored, but 6.5.4 or later support vSphere 4.

@ Ishikawa: I did not see a compatility list doc on the link you gave.. Can you share the compatibility list stating ESX 4 is supported?
