File being locked by another process?

I get the following error when I try to run a backup job on a few of the larger partitions (250gb+). The jobs on my smaller partitions seem to work just fine. This happens on multiple servers. I'm just backing up to an external hard drive, and have tried both USB 2.0 and ESATA connections.

 

I'm running Windows 2003 R2 standard, and have all the latest updates and service packs. I read in the kb that the administrative share created by default might be causing this, so I tried creating a user share and using that. No dice. I'm at a loss as to what's going on. Anyone else happen to have seen this error or have a clue as to what is going on? Any help is appreciated!
