A current consideration for a DR site configuration is to have the Netbackup Master server also running the DNS Server role. Having never considered this previously, would this cause any problems ? DNS and NBU running on the same W2K8R2 server........

It will not cause any issues but make sure you have enough resource available on the master server so it can handle the dns queries and master server database. There is no compatibility issues running dns and nbu on the same maching.
The only concern is if there is some issue in DNS or in Netbackup then for troubleshooting you are making both the applications down. It is recommended in big environments to have the applications run independently on servers.
We had once master server on file server and whenever we had some issues on NBU we had to take downtime with IT service manager and it used to take us 2-3 days to get approval and reboot the server. If you have just NBU then as a admin you are the owner of it and can reboot the server as and when needed.

Keep in mind that it is always recommended to have multiple DNS servers in your environment for that very reason. Should one be down for maintenance your world doesn't stop.

AJ... I cannot stress enough the fact that you should NEVER rely on a single DNS server. ALWAYS have slave DNS servers available so that if a server is down your name service is not affected. It is absolutely ok to use the Netbackup server as a name server just make sure you have additional slave DNS servers in your environment as well.
All the best
